柯尊记,男,1996年于中山大学医学院解剖学与神经生物学专业获博士。先后在中国科学院上海脑研究所,美国伊利偌斯大学医学院解剖学与细胞生物学系和美国康奈尔大学医学院神经科学系从事博士后研究。1998-1999年任郧阳医学院副教授,解剖学教研室主任。2003年底加入中国科学院上海生科院营养科学研究所,2004年入选中国科学院“百人计划”。2006年入选上海市“浦江人才计划”。现任中国科学院上海生命科学研究院营养科学研究所研究员,博士研究生导师,神经营养科学研究组组长。

研究内容

长期从事衰老的神经生物学机制研究,发现了氧化代谢紊乱、氧自由基产生增加、免疫/炎症反应是维生素B1缺乏(TD)引起的选择性中线旁丘脑核神经元死亡的重要病理生理过程;内质网应激、未折叠和错误折叠蛋白累积、caspase-12激活是参与代谢性氧化损伤引起的神经元死亡的主要细胞机制;证实神经元产生的单核细胞趋化因子,是氧化代谢紊乱诱发免疫/炎症反应的开关分子。发现氧化代谢紊乱可以通过增强β分泌酶的活性增加β淀粉样蛋白的产生,从而加速神经元病变的发展过程。TD可增加淀粉样蛋白的产生,亚临床维生素B1缺乏是老年性痴呆的危险因素之一。

研究领域

1)中西医结合防治神经退行性疾病。在传统中药中寻找可以延缓老年性神经退行性疾病发展的生物活性小分子和中药复方。为防治老年性神经退行性疾病,尤其是阿尔茨海默病提供策略和方法。

2)阐明老年性神经元死亡的可能细胞、分子生物学机制,寻找能客观反映老龄健康的生物学标记,和延缓老年性神经元死亡的分子靶点。

3)探讨酒精滥用等环境因素对神经系统发育和衰老影响的细胞、分子生物学机制。
